Study of Mycelial Growth of Cordyceps sinensis on Different Culture Media

In this research work, four growth media: potato dextrose agar (PDA), potato dextrose yeast agar (PDYA), malt extract agar (MEA) and yeast extract agar (YEA) were used for the production of mycelium of Cordyceps sinensis. Mycelium growth of Cordyceps sinensis was the best in PDA media followed by MEA and PDYA media respectively, while it was the lowest for YEA media. To screen out a suitable pH level for the mycelial growth, PDA media was adjusted at different pH level (4.0, 4.5, 5.0, 5.5, 6.0, 6.5, 7.0, 7.5, 8.0, 8.5, 9.0, 9.5 and 10.0) and it was found that alkaline media (8.5-9.5) was better for the mycelial growth of Cordyceps sinensis, although the variation of growth at different pH levels was not significant and mycelial growth occurs at a wide range of pH. To screen out a suitable temperature for the mycelial growth, culture medium was incubated at different temperatures (≤10ºC, 15ºC, 20ºC, 25ºC, 30ºC and 35ºC) and it was found that mycelium growth of Cordyceps sinensis occured at 10-25 ºC and it was best for 20-25 ºC, while no mycelial growth was found at 30 ºC and 35 ºC.
